This is a list of enemies that made a first appearance in a Super Mario game, and were never brought back in any other game, even outside of the Super Mario series. Cameos and remasters of these games don't count as new appearances.

Ant

Super Mario Land 2: 6 Golden Coins These small enemies continuously walk from side-to-side and march into other enemies to deal damage.

Aqua Goomba

Super Mario Land 2: 6 Golden Coins These enemies are Goombas wearing diving helmets. Despite their name and appearance, they generally appear free from the water and behave largely like regular Goombas, with the only difference being that they are not flattened when jumped on.
An Astro-Lanceur in Super Mario Odyssey

Astro-Lanceur

Super Mario Odyssey These enemies are large, flying, silver octopus-like creatures with six arms. They have several spikes protruding from each arm as well. Touching one of these arms will deal one point of damage. They spit cone shell projectiles at Mario as an attack. These shells explode upon contact. They are based off of Mollusque-Lanceur.

Baby Cheep

Super Mario Bros. 3 These enemies are small Cheep-Cheeps that Big Berthas take care of. Big BerthS periodically spit their babies out and put them back in their mouth.

Ball Beamer

Super Mario Galaxy These are mechanical enemies that are immobile. They discharge circular shockwaves that travel around the circumference of the planet, damaging the player if he touches them.

Balloon Boo

New Super Mario Bros. Only appearing in World 4-Ghost House, they are Big Boos with pink cheeks. They appear large at first, but when looked away from, they approach the player while expelling air and shrinking; otherwise, they stop and inflate by inhaling air at a rather slow rate, until they have assumed their massive appearance once more.

Batadon

Super Mario Land These are hopping stone head enemies, similar to a Fighter Fly or Kumo, except it bounces higher, moves slower and does not pause on the ground. To defeat this enemy, the player either has to jump on top of it, or shoot three Superballs at it.

Battle Beetle

Super Mario Land 2: 6 Golden Coins These enemies are flying beetles that simply walk forward until they reach a ledge. When they do, they fly straight up, then slowly hover down at an angle towards the player.

Bear

Super Mario Land 2: 6 Golden Coins These enemies resemble teddy bears and are found riding atop Beach Balls, which they can use to traverse spiky and dangerous floors.

Bee

Super Mario Land 2: 6 Golden Coins These enemies are found near walls with eight holes in them. Each hole houses a different Bee, which will exit its hole after the previous is defeated. They try to attack by exiting their holes and slowly homing in on the player.

Super Mario Land 2: 6 Golden Coins These enemies resemble Japanese paper lanterns, and their attack method involves them sticking out their tongues. They cannot be destroyed, but they can be used as platforms to step on.
Big Eep Cheeps

Big Eep Cheep

New Super Luigi U These enemies are just a large version of Eep Cheeps, and they act just the same.
A Big Grinder

Big Grinder

New Super Mario Bros. 2 These are just large versions of Grinders, and they act the same. They only appear in one level, World 3-Castle, although they're extremely common there.
A Big Poison Piranha Plant in Super Mario Odyssey

Super Mario Odyssey These enemies are just larger variants of Poison Piranha Plants, although they create a larger poison puddle.

Bigbee

Super Mario Land 2: 6 Golden Coins This is a Bee enemy that if the player goes near, it will drop down and then slowly fly back up to its position upon hitting the floor.

Birdfish

Super Mario Land 2: 6 Golden Coins These enemies are similar to Cheep Cheeps, jumping out of a body of water and attacking anything in their way. However, these glide in the air a bit longer than Cheep Cheeps, using their wings to fly through the air.

Bitefrost

Super Mario Odyssey These enemies are in the shape of a large pillar, with the top of their bodies being their eyes and two sets of blocky teeth. They hide underground, showing a shadow of where they are, awaiting for the player to stand directly above them, and they will rise up above ground, carrying the player with them. They will wait a few seconds before gnawing their teeth as an attack.

Super Mario Maker 2 These enemies are a variant of Mechakoopas, being red and pink colored versions that fire homing Bull's-Eye Bill-like missiles from their mouths. These missiles explode shortly after being launched, which are capable of destroying blocks and defeating enemies.

New Super Mario Bros. This type of enemy disguises itself as a ? Block with bricks on top of it, which can be distinguished by the fact that the question mark on it is motionless. Once the player approaches it, it reveals a black body with blue feet. They hop at every vocal sung in the background music, and if the player is standing on top of one, this enemy will move in the direction that he is facing.

Super Mario 3D World These red block-shaped enemies continually march back and forth along a set path. They will always travel in groups of two or more, and players xan defeat them in a group by landing on them with any jump or attack. Defeating them in a group will cause the rest to scatter as they frantically run around, which after a few seconds, they will regain composure and angrily chase after any characters nearby in unison.

Super Mario 3D Land These enemies have pointed green heads covered in spikes and their body is usually composed of Brick Blocks and/or ? Blocks, and the tops of these blocks are also spiked. They do not attack the player but simply hover overhead in a set route. They can easily be avoided by just walking underneath them.
Jumping Blooper

Bloopad

Super Mario Sunshine These enemies occasionally jump out of the water, leaving them susceptible to being sprayed by F.L.U.D.D.. When sprayed, they land on the water and flatten. They can be used as platforms in this condition and thus are useful in helping the player cross slimy water. The player must cross quickly, however, as they revert to their normal form after about fifteen seconds.

Super Mario Sunshine These Koopa-like enemies throw their shells at the player. The shells can electrocute him and remove a portion of his health. The shells return to them like boomerangs.

Super Mario 3D World These enemies are pink, blocky and squishy, and appear in groups and primarily serve as guardians to secret passages. Contact with one damages the player. They can be temporarily disposed of by any normal means of attack except jumps, by striking a nearby gong, or by use of the Wii U gamepad touchscreen.

Bomubomu

Super Mario Land 2: 6 Golden Coins These are pig-like enemies that walk left and right, shooting cannonballs from their cannon-shaped snouts, which are aimed horizontally or diagonally upwards, depending on where the player is when it stops. The player can jump on these cannonballs to destroy them.

Super Mario Galaxy These enemies are metallic-looking fish that have glowing red eyes, spinning, propeller-like fins, and attack in a way that is very similar to Torpedo Teds: by homing in on the player and striking them to cause damage. They can be destroyed by being led to the surface.

Bone Twister

Super Mario Galaxy These tornado-like enemies possess eyes and have debris spinning inside them, such as skulls and bones. They can be helpful at times, being used to traverse obstacles. Touching one also causes them to spin around.

Keep in mind, it's not really fair to compare Super Mario Odyssey to older games as it was the last 3D mainline Super Mario game released, therefore it's harder for enemies that work well in a 3D space to appear in other 2D games. Super Mario Bros. Wonder is not included at all in this list for the obvious reason that it is the last Super Mario game released.
